The Laboratory of Autophagy and Membrane Dynamics at KU Leuven, led by Professor Alex van Vliet, is seeking a highly motivated PhD candidate to join a dynamic research team in the Department of Cellular and Molecular Medicine. The lab focuses on uncovering the molecular mechanisms of autophagy initiation and membrane dynamics, employing cutting-edge techniques such as in vitro reconstitution, structure-function studies, biochemistry, and cell biological approaches. The research aims to advance understanding of how selective autophagy targets and degrades toxic cargo, including protein aggregates and damaged organelles, which is crucial for cellular health and the prevention of neurodegenerative diseases.
As a PhD student, you will investigate the molecular mechanisms by which autophagy machinery recognizes and eliminates toxic aggregates and organelles. The project leverages state-of-the-art technologies, including high-resolution fluorescence microscopy, quantitative proteomics (HDX and XL MS), (cryo-)electron microscopy, and advanced cellular assays utilizing CRISPR/Cas9 genome editing. Initial studies will be conducted in tractable cell lines to elucidate protein complex formation, followed by experiments in iPSC-derived neuronal models and patient samples to translate findings to disease-relevant contexts.

Bonus criteria include experience with molecular and cell biology techniques (cell culture, Western Blotting, PCR/qPCR, cloning, recombinant protein expression/purification), fluorescence microscopy or image analysis software, advanced cell manipulation (CRISPR/Cas9, flow cytometry/FACS), and basic familiarity with data analysis tools (GraphPad Prism, R, or Python).
