Regulation of the deacylase ABHD17 in synaptic plasticity

This PhD project at Coventry University investigates the regulation of the deacylase ABHD17 in synaptic plasticity, focusing on the reversible protein modification process known as S-acylation (palmitoylation). S-acylation is highly prevalent in the brain, regulating nearly half of all synaptic proteins and playing a critical role in neurological disorders, cancer, metabolic, and cardiovascular diseases. The project aims to elucidate how ABHD17 activity is regulated in cultured rat hippocampal neurons during synaptic activation, with a particular emphasis on the postsynaptic scaffold protein PSD-95 and its role in glutamate receptor downregulation and synaptic plasticity. As a PhD student, you will employ a range of advanced techniques including biochemistry, chemical biology, and confocal microscopy. You will gain hands-on experience in preparing rat hippocampal neuronal cell cultures, confocal imaging, molecular biology (PCR, mutagenesis, subcloning), and protein biochemistry (S-acylation assays, click chemistry, SDS-PAGE, western blotting, and co-immunoprecipitation). The research is conducted within an international and multidisciplinary team of cell and molecular biologists, biochemists, and computational chemists. The program is structured as a joint initiative between GITAM Visakhapatnam campus (India) and Coventry University (UK), with Years 1, 3, and 4 based in India and Year 2 at Coventry University. The studentship covers tuition fees, a bursary, one return economy airfare, visa, and overseas healthcare for the mobility period. Comprehensive research training and career development support are provided through Coventry University's Doctoral and Researcher College. Applicants should hold at least a 2:1 first degree in a relevant discipline with a minimum 60% mark in the project element and overall module average. English language proficiency (IELTS 7.0 overall, minimum 6.5 in each component) is required. Previous experience in protein biochemistry and primary cell culture is advantageous.
