About this position
This project is on offer as part of the studentship scheme of the Euan MacDonald Centre for Motor Neuron Disease Research. The Euan MacDonald Centre is a multi-disciplinary network of motor neuron disease (MND) researchers across Scotland. MND is a rapidly progressing, and ultimately fatal, neurodegenerative disease with no cure. Despite important advances in our understanding of the disease, and major medical advances, there are no effective disease modifying treatments. This highlights a clear need for the identification of disease modifying treatments. In this proposed research we have exploited novel computational approaches combined with human stem cell platforms to identify potential re-purposed medicines that can be fast-tracked for testing in clinical trials.
Dysregulated RNA metabolism is a major hallmark of MND. Notably, pathological cytoplasmic TDP43 aggregates leads to nuclear loss-of-function (LOF) of TDP43 which controls splicing of a plethora of genes. In this study, we aim to identify drug repurposing opportunities, either as single or combination therapeutics, that reverse the transcriptomic changes associated with TDP43 pathology in MNs. To undertake this, we will use a state-of-the art technique D igital R NA with pert U rbation of G enes (DRUG-seq) that allows high-throughput drug screening using transcriptomic changes as a readout.
Our approach overcomes the challenges and timescales associated with traditional drug discovery to bring disease modifying therapeutics to MND patients faster.
