About this position

This opening is for a postdoctoral researcher to join the ERC Synergy project 3Stops2go, focused on engineering alternative decoding for therapeutic purposes. The work sits at the intersection of RNA, translation, biochemistry, biophysics, and biological engineering.

The project has two main goals: (1) designing and analysing a large-scale screen in human cells to identify suppressor tRNAs that can efficiently read through premature termination codons while remaining selective for native stop codons; and (2) elucidating the mechanism of action of TLN468 and its derivatives, a readthrough inducer with unusual selectivity. The successful candidate will contribute to quantitative reporter design, functional screening, high-throughput data analysis (NGS and ribosome profiling), and mechanistic interpretation.

Planned activities include molecular biology (cloning, expression, in vitro transcription and translation), biochemical work (protein purification, luciferase assays), human cell culture, lentivirus handling, and development of a lentivirus-based screen for tRNA optimisation. The position is based at I2BC (CEA, CNRS, Université Paris-Saclay) in the Génomique, Structure et Traduction team, with access to sequencing, flow cytometry, mass spectrometry, cryo-EM, and the broader Paris-Saclay research environment.

Eligibility highlights: PhD or equivalent in molecular biology, biochemistry, or a related field; strong expertise in RNA and translation; experience with cloning, human cell culture, flow cytometry, and NGS analysis; lentiviral vectors, functional screens, or ribosome profiling are advantageous.
