About this position
Research Officer in the Glukhova lab at the Walter and Eliza Hall Institute of Medical Research
This role sits in the Immunology Division at the Walter and Eliza Hall Institute of Medical Research and focuses on the biochemistry of Wnt signalling. The project is centered on the Wnt signalosome, a large and heterogeneous membrane protein complex that is central to Wnt pathway function and of interest for drug discovery. The successful candidate will join the Glukhova lab, a multidisciplinary group working at the interface of membrane protein biochemistry and structural biology.
The position is highly experimental and hands-on. You will lead a project to reconstitute, characterise, and stabilise multi-component signalling assemblies for mechanistic and structural investigation. The work involves developing and optimising expression, solubilisation, and purification strategies for challenging membrane complexes, with attention to how detergents influence protein integrity and function. It also involves strategies to stabilise heterogeneous complexes for downstream analysis and preparing samples suitable for structural studies in close collaboration with structural biologists in the group.
The project uses structural interpretation, protein engineering, and biochemical reconstitution to understand how Wnt signalling assemblies are built and maintained. The role also includes contributing to manuscripts and grant applications, maintaining rigorous records, and helping support the laboratory’s collaborative research environment.
Wehi is a pre-eminent biomedical research institute with a strong research culture, and this position offers the opportunity to work in a multidisciplinary setting spanning biochemistry, structural biology, and pharmacology.

What's required
A PhD in biochemistry, membrane biology, structural biology, or a closely related discipline is essential. Candidates should have demonstrated expertise in expression, purification, and biophysical characterisation of membrane proteins and large multi-subunit protein complexes, practical knowledge of detergent chemistry for membrane protein biochemistry, and experience stabilising heterogeneous or transient complexes using point mutations, conformation-selective binders, or dominant-negative accessory subunits. Strong ability to access, interpret, and critically evaluate Protein Data Bank structures is required, along with strong molecular biology skills including construct design, cloning, and mutagenesis. Experience with mammalian and/or insect cell expression systems is required, and applicants should have excellent written and verbal communication skills with a track record of peer-reviewed publications. Desirable experience includes X-ray crystallography or cryo-EM workflows, biophysical techniques for complex assembly and heterogeneity, FACS, stable cell line generation or CRISPR editing, fluorescence- or luminescence-based pharmacology or binding assays, and computational structural tools such as PyMOL, Chimera, or AlphaFold.
