About this position
PhD studentship at the University of Exeter on the functional genomic characterisation of genetic risk for diabetic neuropathy using human stem cell models.
This project addresses a major complication of diabetes that affects peripheral sensory nerves and currently has no treatment that halts or reverses disease progression. The research focuses on APOE, a genetic risk factor with links to neuropathy severity, and asks whether risk is driven by sensory neurons, Schwann cells, or their interaction within the neuron-glia unit.
The successful student will build and analyse human sensory neuron and Schwann cell co-culture systems from stem cells carrying APOE e2/e2, e3/e3, or e4/e4 on a shared genetic background. Experiments will test cell survival, nerve fibre integrity, ensheathment, and network activity under high glucose and cytokine challenge. The project will also use mixed co-cultures to separate neuronal from glial effects, and long-read RNA sequencing to identify transcriptomic and isoform-level changes associated with risk. Functional follow-up will help prioritise candidate genes and pathways that may be druggable.
Methods and training include stem cell biology, genome editing, microelectrode array analysis, patch clamp electrophysiology, apoptosis assays, high-content imaging, super-resolution microscopy, Oxford Nanopore sequencing, transcriptomics, and statistical modelling. The project is interdisciplinary and spans three institutions plus industrial input, with existing collaborations providing isogenic APOE lines and protocols for generating sensory neurons.
Facilities at Exeter include high-content imaging, super-resolution microscopy, and Oxford Nanopore sequencing. The funding notes state that the studentship provides fees plus a stipend of £21,805 per annum for 2026/7 and is funded by the EPSRC. The application deadline is 8 December 2026.
This is a strong opportunity for a student interested in neurodegeneration, diabetes complications, human cell models, functional genomics, and translational biomedical research.
