About
Achim Lother is an Attending Physician and Research Group Leader at the University of Freiburg, Germany. His research focuses on cardiovascular diseases, particularly the molecular mechanisms underlying heart failure, myocardial injury, and the role of various cellular components in cardiac health. Recent articles authored by him explore topics such as endothelial cell cycle regulation, cardiac myopathy, and the pharmacology of mineralocorticoid receptor antagonists.

PhD position available in Cardiovascular Epigenetics at the University of Freiburg within the FOR6051 programme, The Interstitium: a Key Determinant of Cardiac Function . The project (P6-2) is hosted at the Institute for Experimental and Clinical Pharmacology and Toxicology and is supervised by Achim Lother with a joint project involving Rémi Peyronnet . The research investigates how altered hemodynamics in heart failure and atrial fibrillation affect epigenetic regulation of gene expression, with a focus on mechanosensitive ion channels , cardiac remodeling, and arrhythmogenesis. Research activities include translational cardiac disease models, pharmacology, molecular biology, wet-lab omics technologies, and bioinformatics. The project mentions in vivo experimental research, cell isolation, flow cytometry, transcriptomics, epigenetics, RNA-seq, ChIP-seq, ATAC-seq, and proteomics with bioinformatics analysis. Eligible backgrounds include (molecular) medicine, biochemistry, (technical) biology, pharmacy, and bioinformatics. Applicants should have a strong motivation, a solid background in cardiac biology and/or epigenetics, and preferably prior bioinformatics experience. English proficiency at B2 or higher is required. The application window is open until 07 June 2026 . Applications are submitted via the SGBM portal under the FOR6051 track. Required documents include personal/contact details, bachelor and master transcripts/certificates, proof of English proficiency, contact details of two referees, and a motivation statement with research interests.

Endothelial cells play crucial roles in physiology and are increasingly recognized as therapeutic targets in cardiovascular disease. Here, we analyzed the regulatory landscape of cardiac endothelial cells by assessing chromatin accessibility, histone modifications, and 3D chromatin organization and confirmed the functional relevance of enhancer-promoter interactions by CRISPRi-mediated enhancer silencing. We used this dataset to explore mechanisms of transcriptional regulation in cardiovascular disease and compared six different experimental models of heart failure, hypertension, or diabetes. Enhancers that regulate gene expression in diseased endothelial cells were enriched with binding sites for a distinct set of transcription factors, including the mineralocorticoid receptor (MR), a known drug target in heart failure and hypertension. For proof of concept, we applied endothelial cell–specific MR deletion in mice to confirm MR-dependent gene expression and predicted direct MR target genes. Overall, we have compiled here a comprehensive atlas of cardiac endothelial cell enhancer elements that provides insight into the role of transcription factors in cardiovascular disease.
